From April 2000 to July 2001, at the Wilmer Eye Institute, Johns Hopkins University School of Medicine, USA, under the mentorship of Professor Cao Anmin, a world-renowned ophthalmic pathologist, I conducted postdoctoral research on the molecular biological mechanisms of retinal neuronal apoptosis. Retinal neuronal degenerative diseases (such as age-related macular degeneration, retinitis pigmentosa, and chronic simple glaucoma) are major blinding ophthalmic conditions, with apoptosis being the common final pathway of cell death. Currently, there are no effective curative treatments for these diseases in clinical practice, primarily due to the unclear molecular biological regulatory mechanisms of apoptosis. Using molecular biology experimental techniques, I focused on investigating the regulatory mechanisms of the transcription factor NF-kB on retinal neuronal (photoreceptor and ganglion cell) apoptosis, exploring the possibility of blocking neuronal apoptosis, which holds significant importance. The research findings, titled "Increased Nuclear Factor-kB p65 Immunoreactivity Following Retinal Ischemia and Reperfusion Injury in Mice," were officially published in the Journal of Neuroscience Research (USA) in 2003 (IF 3.239) and received the Bronze Award for Outstanding Paper at the 3rd National Youth Ophthalmology Academic Conference. ② Studies on corneal wound healing mechanisms following excimer laser photorefractive keratectomy for myopia. This research primarily investigated the mechanisms of subepithelial corneal haze formation after PRK for myopia, as well as pharmacological preventive measures, to further improve surgical outcomes. The findings have been published in core journals of the Chinese Journal of Ophthalmology. ③ Studies on the mechanisms of myopia onset and progression. Myopia is a common ophthalmic condition whose onset and progression mechanisms remain unclear, and there are currently no clinical methods to fundamentally halt its progression. We established animal models of myopia and utilized molecular biology and immunohistochemical techniques to study the mechanisms of axial myopia onset and progression, aiming to explore novel therapeutic approaches to prevent the onset and progression of myopia.
